Abstract:
This study was carried out to isolate and identify the protein and to compare the expressed proteins among ripening stage by a protein profile with different functional properties using two-dimensional electrophoresis (2-DE) technique. In these studies, total number of detected protein spot was 1,384 and well-defined 29 spots could be resolved on the 15th day after silking day. In the expressed protein spots according silking stages, they could detect well-defined 35(20th days after silking stage), 45(25th days after silking stage) and 47(30th days after silking stage) spots, respectively. Among 43 identified protein spots used by two-dimensional electrophoresis, 10 different protein spots were found in multiple spots which were globulin-2 and vicilin-like embryo storage protein. Three spots were identified to uncharacterized protein. Globulin-2 in multiple spots was of the same pI and molecular mass. The largest functional category was that of stress response proteins. Second, the number of protein in the regulatory protein category was large. As protein spots was classified as molecule function, purple corn Heukjinjuchal kernel was isolated by five functional categories such as regulatory protein, stress response, signal transduction, metabolism and cell structure except for the unknown protein. The most abundant molecular function category in purple corn was cell structure (34.4%) which have a main role in storage protein. The next abundant categories of purple corn kernel were stress response (18.8%), metabolism (15.6%), regulatory protein (12.5%) and signal transduction (3.1%).
